High Throughput Cell Adhesion: Screening of VLA-4 antagonists under physiological shear flow conditions

Cell adhesion governs countless biological phenomena in health and disease. Protein- protein or protein-carbohydrate interactions plays an important role in directing cell migra- tion, gene expression, morphological changes, and programmed cell death. Typical molecules involved in cell-cell and cell-matrix adhesion are integrins, cadherins, cell adhesion molecules (-CAMs) and ADAMs (transmembrane proteins with disintegrin and metalloprotease domains) (Yamada, KM 2003). Cell adhesion molecules can be co-opted by other cell types in the body contributing to inflammation and metastatic diseases. The molecular interactions governing adhesion between white blood cells and cancer cells and the cells of the endothelium is the focus of targeted therapeutic drug development.
